چکیده

We investigated the activation of the nonenzymatic protein cofactors factor VIII and factor V in plasma when coagulation was initiated by thromboplastin. With sensitive bioassays, we were able to measure specifically the generation of activated factor VIII and activated factor V in plasma. Our results showed that when plasma was triggered with a relatively high concentration of thromboplastin, factor VIII and factor V were completely activated at the clotting time of plasma. However, when the generation of thrombin, but not that of factor Xa. was delayed by addition of hirudin to

Evaluation of the initiation phase of blood coagulation using ultrasensitive assays for serine proteases.

The initiation phase of enzyme generation in a reconstituted model of the tissue factor (TF) pathway to thrombin was evaluated. At 1.25 pM added TF, no thrombin generation was observed in the absence of factor V. The substitution of factor Va for factor V increased the rate of thrombin generation. Factor X activation during the initiation phase was not influenced by the absence of factor VIII o...

Activated factor V is a cofactor for the activation of factor XI by thrombin in plasma.

The mechanism by which the intrinsic pathway of coagulation contributes to physiological hemostasis is enigmatic. Thrombin activates factor XI, a key zymogen in this pathway, which leads to increased thrombin generation. As thrombin-dependent activation of factor XI in vitro is relatively inefficient, we hypothesized that a physiological cofactor supports this reaction in a plasma environment. ...

Importance of factor-IX-dependent prothrombinase formation--the Josso pathway--in clotting plasma.

We report a study on the importance of factor IX activation in thromboplastin-dependent coagulation in plasma. Diluted, CaCl2-containing thromboplastin solutions at constant phospholipid concentration were used to trigger the coagulation in plasma from patients with congenital factor IX and factor VIII deficiency in the presence and absence of added factors IX and VIII, and the generation of th...

The importance of activation of antihemophilic globulin and proaccelerin by traces of thrombin in the generation of intrinsic prothrombinase activity.

seconds.2 Therefore, although the PTT measures the over-all time of the intrinsic clotting reactions, in most circumstances it essentially measures the rate of generation of intrinsic prothrombinase. It is now generally accepted that contact with a negatively charged surface such as glass initiates clotting in vitro by activating Hageman factor ( HF, factor XII ) . Activated HF then reacts with...
